July 15, 2010: "NASA-funded researchers are monitoring a big event in our planet's atmosphere. High above Earth's surface where the atmosphere meets space, a rarefied layer of gas called "the thermosphere" recently collapsed and now is rebounding again."

"This is the biggest contraction of the thermosphere in at least 43 years," says John Emmert of the Naval Research Lab, lead author of a paper announcing the finding in the June 19th issue of the Geophysical Research Letters (GRL). "It's a Space Age record."

"The collapse happened during the deep solar minimum of 2008-2009—a fact which comes as little surprise to researchers. The thermosphere always cools and contracts when solar activity is low. In this case, however, the magnitude of the collapse was two to three times greater than low solar activity could explain. "Something is going on that we do not understand," says Emmert."

[(11)C]P943 is a new radioligand recently developed to image and quantify serotonin 5-Hydroxytryptamine (5-HT(1B)) receptors with positron emission tomography (PET). The purpose of this study was to evaluate [(11)C]P943 for this application in humans, and to determine the most suitable quantification method. Positron emission tomography data and arterial input function measurements were acquired in a cohort of 32 human subjects. Using arterial input functions, compartmental modeling, the Logan graphical analysis, and the multilinear method MA1 were tested. Both the two tissue-compartment model and MA1 provided good fits of the PET data and reliable distribution volume estimates. Using the cerebellum as a reference region, BP(ND) binding potential estimates were computed. [(11)C]P943 BP(ND) estimates were significantly correlated with in vitro measurements of the density of 5-HT(1B) receptors, with highest values in the occipital cortex and pallidum. To evaluate noninvasive methods, two- and three-parameter graphical analyses, Simplified Reference Tissue Models (SRTM and SRTM2), and Multilinear Reference Tissue Models (MRTM and MRTM2) were tested. The MRTM2 model provided the best correlation with MA1 binding-potential estimates. Parametric images of the volume of distribution or binding potential of [(11)C]P943 could be computed using both MA1 and MRTM2. Methods: Studies were performed in 3 isoflurane-anesthetized rhesus monkeys with 3 bolus+infusion deliveries of [11C]P943 per day on 1-2 days. Specific activity (SA) was varied from 2-200 MBq/nmol. Dynamic scans were acquired on the HRRT and 8 ROIs were defined on MR images co-registered to summed PET images. Binding potentials (BPND), determined by the equilibrium ratios of ROIs to cerebellum (CER), were used to estimate Bmax and Kdapp (Kd/fND) by non-linear fitting. Two analyses were performed: 1) assuming NDU was equal to CER activity, and 2) estimating NDU by a scale factor (S) times CER activity.
